Quantitativer Nachweis des "Spice"-Wirkstoffes JWH-018 im Serum mittels Fluessigchromatographie-Tandemmassenspektrometrie / Quantitative detection of the active "Spice" ingredient JWH-018 in serum by means of liquid chromatography-tandem mass spectrometry

Nach dem Rauchen der Kraeutermischung "Spice" berichteten Konsumenten in einschlaegigen Internetforen von einer halluzinogenen Wirkung. Sieben verschiedene Raeuchermischungen wurden mit GC-MS und LC-MSIMS auf die fuer den psychogenen Effekt verantwortlichen synthetischen Cannabinoide untersucht. Die Raeuchermischungen enthielten ein CP47,497-C8-Homologes und JWH-018 entweder einzeln oder beide Komponenten zusammen. In einigen Mischungen konnte keines der synthetischen Cannabinoide nachgewiesen werden. Nach StGB wird sanktioniert, wer infolge von Genuss berauschender Mittel nicht in der Lage ist, ein Fahrzeug sicher zu fuehren. Grundlage fuer die Sanktionierung ist somit der analytische Nachweis der psychoaktiven Substanzen im Blut des Fahrers. Da die verschiedenen "Spice"-Raeuchermischungen JWH-018 zum Teil als alleinige Wirkkomponente enthalten, wurde eine Methode zum quantitativen Nachweis von JWH-018 im Serum entwickelt. Im Einzelnen wurden die Proben auf C18-Festphasenextraktionssaeulen aufkonzentriert und die Extrakte effektiv am LC-MS/MS-System analysiert. Die Nachweisgrenze und Bestimmungsgrenze des Verfahrens ergaben sich zu 0,2 ng/mL und 0,6 ng/mL. Andere gaengige Drogen beeintraechtigten die Quantifizierung von JWH-018 im Serum nicht. Bei Anwendung der Nachweismethode auf forensische Blutproben wurden JWH-018 Konzentrationen von 0,2 ng/mL bis 2,5 ng/mL gemessen. Somit gelingt der fuer die Strafverfolgung essentielle analytische Nachweis von JWH-018 als einer der psychoaktiven lnhaltsstoffe der Droge "Spice". (A) ABSTRACT IN ENGLISH: In a number of relevant internet forums, consumers have reported a hallucinogenic effect from smoking the incense drug "spice". Seven incense mixtures were analyzed using GC-MS and LC-MS/MS in order to test for synthetic cannabinoids that cause psychogenic effects. The incense mixtures contained either a CP47,497-C8-homologe or JWH-018, or both in combination. Synthetic cannabinoids could not be detected in some mixtures. The German penal code sanctions persons who drive a motor vehicle unsafely following consumption of inebriating substances. Therefore the legal basis of the sanction is analytical proof of psychoactive substances in the driver's blood. A method for quantitative detection of JWH-018 in serum was developed because some of the "spice" incense mixtures contained JWH-018 as the only active component. More specifically, serum samples were concentrated using a C18 solid-phase extraction column and the extracts were effectively analyzed using the LC-MS/MS System. The detection limit and the quantification limit were 0.2 ng/mL and 0.6 ng/mL respectively. Other popular drugs did not interfere with this highly selective quantification of JWH-018 in serum. When using this detection method to test forensic blood samples, JWH-018 concentrations ranging from 0.2 ng/mL to 2.5 ng/mL were measured. Therefore it is possible to detect JWH-018 - one of the psychoactive ingredients of the incense drug "spice"- in serum and thus provide the analytical evidence necessary for criminal prosecution.
