LOW LIGHT LEVEL LUMINESCENCE; THE VIDEO FLUOROMETER: A NEW INSTRUMENT FOR MULTICOMPONENT ANALYSIS

The video fluorometer which uses a low light-level television camera as a multichannel photometer, acquiring excitation and emission spectra simultaneously is described. The design of the apparatus is based on the use of polychromatic illumination of the sample. The excitation beam is produced by focusing the image of a 150-W xenon arc lamp on the entrance slit of a monochromator mounted with the long axis of the entrance slit perpendicular to the long axis of the sample cuvet. The exit slit of the monochromator is removed, and the spectrally dispersed continuum of radiation present at the exit slit plane is re-imaged onto the sample. Details are outlined of the analysis in which the fluorescent substances which absorb light most strongly in the red region of the spectrum will fluoresce most strongly near the top of the cuvet, while those which absorb most strongly in the blue will floresce most strongly near the bottom of the cuvet. A number of tests were performed to assess the performance of the video fluormeter and the sensitivity of the system. Applications for the video fluorometer are as numerous as the present applications for conventional fluorometry. One noteworthy application is in the identification of oil spills.
